The Bioavailability Problem with Magnesium Oxide

Magnesium oxide is the most common form of magnesium in budget supplements — it’s cheap, dense (meaning small pills can contain high stated amounts), and easy to manufacture. Its primary problem is poor bioavailability: multiple studies have demonstrated that magnesium oxide is absorbed at approximately 4% efficiency from the gut.* A supplement stating “400mg magnesium (as magnesium oxide)” may deliver fewer than 20mg of usable magnesium to the bloodstream.*

The osmotic effect of unabsorbed magnesium in the colon is what causes the laxative effect associated with magnesium oxide at high doses — it draws water into the gut and accelerates transit.* This is occasionally useful clinically, but it’s not a desirable property in a daily magnesium supplement.*

Why Chelated Magnesium Absorbs Better

Chelation refers to the bonding of a mineral ion to an organic molecule — in the case of magnesium glycinate, magnesium is bound to two molecules of glycine (an amino acid). This chelation changes the absorption mechanism:*

  • Amino acid transporter pathway: Chelated minerals are absorbed through amino acid transporters in the intestinal wall — a different pathway than the mineral ion channels that ionic minerals (like magnesium oxide) must use.* Amino acid transporters have higher capacity and are less saturable than mineral channels.*
  • Reduced competition: Ionic magnesium competes for absorption with calcium, zinc, and other divalent cations.* Chelated magnesium, absorbed as an amino acid complex, largely bypasses this competition.*
  • Stability in the GI environment: Chelated magnesium resists being broken down by stomach acid or binding to dietary phytates and oxalates that can sequester ionic minerals in the gut.*

The result is substantially better absorption efficiency — multiple studies have demonstrated that glycinate and bisglycinate forms of magnesium are significantly better absorbed than oxide forms, with lower rates of GI side effects.*

Glycine’s Additional Properties

Glycine — the amino acid in magnesium glycinate — is not biologically inert. It is an inhibitory neurotransmitter in the central nervous system, a substrate for glutathione synthesis, and has been studied independently for its role in sleep quality.* Bannai et al. (2012, Sleep and Biological Rhythms) found that oral glycine supplementation improved subjective sleep quality and reduced daytime sleepiness in people with sleep complaints.* This means magnesium glycinate provides both the mineral and an amino acid with its own studied biological properties.*

At the cellular level, magnesium functions as:*

  • A cofactor for ATP synthesis — essentially all ATP in the cell exists as Mg-ATP, meaning magnesium is required for every energy-producing reaction*
  • A regulator of calcium channels — magnesium naturally antagonizes calcium entry into cells, supporting normal muscle relaxation after contraction*
  • A cofactor for DNA polymerase and RNA polymerase — required for DNA replication and RNA synthesis*
  • A structural component of ribosomes — required for protein synthesis*
  • A regulator of neuromuscular transmission — magnesium modulates the release of neurotransmitters at the neuromuscular junction*
