What Purity Means Technically

In analytical chemistry and pharmaceutical science, purity refers to the percentage of a substance in a sample that is the target compound — in this case, melatonin. A “pure” melatonin powder would ideally be 100% melatonin with no contaminants, byproducts of synthesis, or residual solvents from the manufacturing process.

In practice, “pharmaceutical grade” melatonin powder is typically specified at 99%+ purity — meaning less than 1% of the material by weight is anything other than melatonin. This is verified through analytical testing methods including High Performance Liquid Chromatography (HPLC), which can identify and quantify specific compounds in a sample.

What Can Reduce Purity

Several things can reduce the purity of a melatonin powder product:

  • Synthesis byproducts: The chemical synthesis of melatonin produces intermediate compounds that must be separated from the final product. Inadequate purification leaves residual impurities in the finished powder.
  • Residual solvents: Synthesis uses organic solvents that must be removed through drying and purification steps. Residual solvents above acceptable limits are an identity and safety concern.
  • Degradation products: Melatonin is sensitive to light, heat, and moisture. Improper storage or handling can cause degradation — breaking down melatonin into related compounds that reduce the concentration of active melatonin in the product.
  • Adulteration: In unregulated markets, products labeled as melatonin may contain different amounts than stated or, in extreme cases, different compounds entirely. This is the issue documented in the 2017 JCSM study, which found deviations of 83% below to 478% above stated label amounts in commercial melatonin products.

How to Verify Purity When Buying

  • Certificate of Analysis (COA): A legitimate COA from an independent third-party laboratory shows the specific test results for identity, purity, and potency for a given batch. Request one — any manufacturer with nothing to hide provides it.
  • HPLC testing confirmation: The COA should specify the testing method. HPLC is the standard for melatonin purity testing — look for it specifically.
  • Batch number traceability: The COA should reference a specific batch number that matches your product. Generic COAs not tied to a specific lot number are not meaningful verification.
  • cGMP manufacturing: 21 CFR Part 111 compliance requires finished product testing before release. Manufacturers operating under cGMP are required to have quality systems that produce these documents routinely.
